Résumé / Abstract : Les trois protéines d'enveloppe S-AgHBs, M-AgHBs et L-AgHBs du virus de l'hépatite B (HBV). synthétisées à partir d'une unique ORF via trois AUG en phase, ont une extrémité C-terminale commune, mais diffèrent par leur extension N-terminale. La protéine S-AgHBs est constituée du domaine S; la protéine M-AgHBs des domaines S et preS2; et la protéine L-AgHBs des domaines S, preSZ et preSL Le caractère infectieux HBV dépend des 75 résidus N-terminaux de preSl de la protéine L-AgHBs et d'un second déterminant porté par la boucle antigénique (BAG) du domaine S des protéines. Au cours de notre étude nous avons établi la cartographie précise du déterminant d'infectiosité de la.BAG, confirmé son association avec le déterminant majeur d'antigénicité "a", également porté par la BAG et identifié plusieurs épitopes du déterminant "a", reconnus par des anticorps monoclonaux neutralisants. Ces données suggèrent une implication du déterminant "a" à l'étape d'entrée HBV dans les hépatocytes. Enfin, nous avons montré que la BAG était suffisante pour médier l'attachement des virions aux heparans sulfate (HS) des hépatocytes. Cet attachement nécessite les résidus positivement chargés R122 et K141, et dépend de la charge globale de la BAG à la surface des virions. Nos résultats suggèrent également une seconde fonction du déterminant "a", dans urte étape qui suit l'attachement.
Résumé / Abstract : The three HBV envelope proteins produced from a unique ORF via three AUG in frame, share the same C-terminal end, but differ in their N-terminal extension. S-HBsAg protein is made of the S domain only; M-HBsAg is made of the S and preS2 domains; and L-HBsAg is made of the S, preS2 and preSl domains. The HBV infectivity depends upon the N-terminal 75 amino acids residues of the L-HBsAg preS1 domain and a second determinant borne by the antigenic loop (AGL) of the S domain. During our study we established a précise mapping of the AGL infectivity determinant, we confirmed its strong relation with the imunodominant "a" déterminant, also borne by the AGL, and we identified several epitopes of the "a" determinant, recognized by neutralizing monoclonal antibodies. These data suggest an involvement of the "a" determinant at the HBV viral entry. Furthermore, we showed that the AGL was sufficient to mediate the heparans sulfate (HS) binding of virions at the hepatocytes surface. This binding required the positively-charged residues, R122 and K141, and depends upon the global charge of the AGL at the virions surface. Our results suggest another function of the "a" determinant, at a post-binding step.
