Résumé / Abstract : L’urate oxydase recombinante est une protéine utilisée pour le traitement de l’hyperuricémie. Après purification, par chromatographie, elle peut être cristallisée. Quels paramètres gouvernent sa solubilité ? La cristallisation peut-elle avoir lieu directement dans l’extrait brut du microorganisme recombinant et être un autre moyen de purifier cette protéine ? Nous avons mesuré les variations de solubilité et les interactions en solution de l’urate oxydase en fonction de la température, du pH, des sels et des polymères. Cette exploration a conduit à approfondir l’effet d’un surfactant, le poloxamère 188, sur la cristallisation de cette protéine. Les résultats obtenus sur la protéine pure nous ont permis de comprendre quels paramètres physico-chimiques stabilisent cette protéine en solution. Nous avons pu identifier des agents cristallisants compatibles avec un procédé de purification, que nous avons testés sur des solutions impures.
Résumé / Abstract : Recombinant urate oxidase is a protein used for the treatment of hyperuricemia. It can be crystallized once purified by chromatography. Which parameters control its solubility? Can crystallization happen in the crude extract from the recombinant microorganism and be another way to purify this protein? We measured urate oxidase solubility variations and interactions in solution as a function of temperature, pH, salts and polymers. This screening led us to go into detail about the effect of a surfactant, the poloxamer 188, on protein crystallization. These studies enable to understand which physico-chemical parameters stabilize this protein in solution. We also identified crystallizing agents which are compatible with a purification process and tried them on impure solutions.
