Résumé / Abstract : L'osteosarcome, une tumeur osseuse maligne, présente un fort taux de résistance aux traitements et de fréquentes métastases. Notre objectif est de caractériser le rôle du syndécan-2, un protéoglycane transmembranaire, dans les cellules d'ostéosarcome. Nous avons montré le rôle proapoptotique du syndécan-2 dans les cellules ostéosarcomateuses, l'implication de la voie JNK/BAX, et le rôle de la PKC delta. Nous avons montré un défaut de Syndécan-2 dans les tissus ostéosarcomateux, une corrélation entre l'expression du syndécan-2 et l'apoptose in vivo, et l'implication du syndécan-2 dans la réponse aux chimiothérapies. Le syndécan-2 apparaît donc comme une molécule clé de la signalisation apoptotique. Nous avons mis en évidence un modèle d'induction d'apoptose dans les cellules d'ostéosarcomes en surexprimant le syndécan-2. L'identification des mécanismes impliqués dans l'apoptose servira à caractériser de nouvelles cibles thérapeutiques pour améliorer les traitements de l'ostéosarcome.
Résumé / Abstract : Osteosarcoma is a primary malignant bone tumor with frequent metastases. We studied the role of syndecan-2, a transmembrane heparansulfate proteoglycan, in osteosarcoma cells. The aim of this thesis was to characterize the role of syndecan-2 in osteosarcoma. First, we showed the proapoptic role of syndecan-2, involving the JNK/BAX signaling pathway. Then we showed how PKC delta is involved in the apoptotic signaling induced by syndecan-2. Finally, we showed a defect of syndecan-2 in osteosarcoma tissues, a correlation between syndecan-2 expression and in vivo apoptosis, and the involvement of syndecan-2 in the answer to chemotherapeutic drugs. In conclusion syndecan-2 appears as a key molecule of the apoptotic pathway. We identify a model of apoptosis induction in osteosarcoma cells by overexpressing syndecan-2. Afterwards, this model could serve for identifying new therapeutic targets to improve the treatments of osteosarcoma.
