Résumé / Abstract : Les E. coli d'adhésion diffuse (DAEC) sont impliqués dans les infections urinaires et dans les diarrhées. Le seul facteur de virulence identifié est une famille d'adhésines, Afa/Dr, liant le Decay Accelerating Factor (DAF) et le Carcinoembryonic Antigen (CEA) qui appartient à la famille des CEACAM. Notre objectif a été d'étudier les interactions entre les CEACAMs et les DAEC Afa/Dr. Nous avons montré que le le CEACAM1 et le CEACAM6 sont des récepteurs pour les DAEC Afa/Dr et sont recrutés autour des bactéries comme le GM1 et la cavéoline, marqueurs de rafts lipidiques. L'adhésion aux DAF, CEA et CEACAM6 induit la formation de prolongements cellulaires autour des bactéries et nécessite un remaniement du réseau d'actine et des rafts lipidiques, l'intervention des Rho GTPases et la phophorylation des protéines ERM. Nous avons aussi démontré que l'internalisation des bactéries s'effectuait par un mécanisme de type «zipper», indépendant de l'actine et faisant intervenir le CEA et le CEACAM6.
Résumé / Abstract : Diffusely adhering E. coli (DAEC) are involved in urinary tarct infections and diarrhoeas. The only virulence factor identified is a family of adhesines, Afa/Dr, binding Decay Accelerating Factor (DAF) and Carcicoembryonic Antigen (CEA) which belongs to the CEACAM family. Our objective was to study the interactions between CEACAMs and the Afa/Dr DAEC. we observed that CEACAM1 and CEACAM6 are receptors for the Afa/Dr DAEC and are recruited around adhering bacteria as well as the GM1 and the caveoline, two lipid rafts markers. Adhesion to the DAF, CEA and CEACAM6 induces the formation of cellular prolongations around the bacteria and requires a reorganization of the actin network and the lipids rafts, as well as Rho GTPases and ERM proteins phophorylation. We also showed that the internalization of the bacteria was carried out by azipper like mechanism, indenpendently of actin. Moreover, CEA and the CEACAM6 seem to support entered of the bacteria into the cells.
