Résumé / Abstract : Le contexte de la thèse est celui de la biologie structurale, discipline pour laquelle l'un des principaux enjeux consiste à obtenir des informations sur la fonction biologique donc sur la conformation de certaines protéines essentielles dans les sciences du vivant. Une nouvelle technique de cristallisation en 2-D de protéines à une interface eau/air est proposée. Un écoulement organisateur dans la sous-phase aqueuse est suggéré, qui devrait permettre de faciliter les conditions de croissance de monocristaux de grande taille dans l'interface. Un dispositif original est développé conjointement avec un modèle mathématique de l'écoulement afin de définir la géométrie la plus optimale.
Résumé / Abstract : The context of the thesis is that of the structural biology. For this scientific field, one of the principal stakes consists in obtaining information on the biogical function of proteins essential in science. As a consequence, identification of the structure of these proteins from crystals stands as a main goal. For the proteins which do not crystallise in 3-D, it is useful to develop an alternative solution. A new technique for the 2-D crystallization of proteins at an air/water interface is thus proposed. An organizing flow within the aqueous subphase is suggested, which should enhance conditions for the growth of large monocrystals in the interface. An original device is jointly developed with a mathematical model of the organizing flow so as to define the most optimal geometry.
