Résumé / Abstract : Les chaines des récepteurs à l'antigène, qui portent la diversité du répertoire immunitaire, sont formés par le réarrangement des segments V, D, et J. Les mécanismes qui gouvernent ces réarrangements sont encore mal connus et reposent sur l'accessibilité aux séquences qui réarrangent. Parmi les différents éléments qui gouvernent cet accès, nous avons étudié le rôle de l'élément génique TEA, situé en amont immédiat du locus TCR J alpha. La délétion de cet élément entraîne l'absence d'utilisation par les lymphocytes des premiers segments Jalpha.
Résumé / Abstract : The diversity of antigen receptors is composed from the recombination of their V, D and J segments. The mecanisms underlying the regulation of the recombination are poorly understood, but rely on the accessibility of the DNA to the V(D)J recombinase. We showed in this work that the two T cell receptors TCR J alpha are not randomly, but rather coincidentally rearranged in a given T cell. This coincidence relies, in part, on the presence of <<T early alpha>> (TEA), a cis regulatorygenetic element located upstream of the TCRJA cluster.
